Setelah penaklukan Aztek oleh Spanyol, Doña Isabel diakui sebagai keturunan Moctezuma yang sah dan menjadi salah satu penduduk asli Meksiko yang diberi hak encomienda. Penduduk asli lain yang juga mendapatkan hak tersebut adalah saudara tirinya Marina (atau Leonor) Moctezuma dan Juan Sánchez, seorang gubernur di Oaxaca.[1]
