p53 (bahasa Inggris: protein 53, p53) adalah faktor transkripsi yang mendapat julukan dalam bahasa Inggris sebagai "the guardian of the genome", atau "guardian angel gene", dan "master watchman" karena peran protein tersebut dalam menjaga sel dari mutasi genetik akibat kerusakan DNA.
p53 memiliki ekspresi yang beragam seperti berbagai fungsi seluler, regulasi, apoptosis, replikasi DNA, proliferasi, dll. Dalam kondisi normal, jumlah konsentrasi p53 dalam sitoplasma sangat sedikit dan hanya teraktivasi saat sel mengalami stress.[1]
Rendahnya asupan asam folat dan vitamin B6 dapat meningkatkan risiko mutasi genetik pada p53 dan menginduksi kanker usus besar.[2] Upregulasi p53 yang berlebihan, biasanya terjadi pada kasus kemoterapi kanker dengan penggunaan cisplatin, dapat meracuni ginjal, dan perlu diturunkan segera, misalnya dengan penggunaan pifithrin-α.[3]
