Jupe memulai karir aktingnya pada tahun 2015/2016 dengan tampil di serial televisi Penny Dreadful dan Downton Abbey.[4] Kemudian pada tahun 2016, ia memainkan peran penting dalam seri thriller mata-mata The Night Manager bersama dengan Tom Hiddleston.[4] Pada tahun 2016, ia juga memainkan peran penting dalam seri Houdini & Doyle.[5]

Pada tahun 2018, ia membintangi film fiksi ilmiah The Titan,[8][5][10] dan bersama Emily Blunt dan John Krasinski dalam film horor A Quiet Place.[11] Krasinski, yang menjadi penulis dan sutradara film ini, memilih Jupe atas rekomendasi Clooney.[12][13]

Pada tahun 2019, Jupe membintangi film Ford v Ferrari bersama Matt Damon dan Christian Bale.[14] Ia memainkan peran utama dalam film Honey Boy bersama Shia LaBeouf dan Lucas Hedges, sebuah film independen berdasarkan kehidupan pribadi LaBeouf, Jupe memerankan LaBeouf di masa kecilnya.[15][16][17]
