MotoGP musim 2020, akan menjadi musim ke-72 Kejuaraan Dunia Sepeda Motor FIM.
Marc Márquez akan memulai musim sebagai Juara Dunia bertahan, setelah mengamankan gelar kedelapan secara keseluruhan di Buriram.

Pergantian Pembalap
- Johann Zarco dan Red Bull KTM Factory Racing mengumumkan di pertengahan musim 2019 bahwa mereka akan mengakhiri hubungan mereka sebelum waktunya. Zarco awalnya dikontrak sampai akhir 2020.[26] Tempat duduknya diambil oleh juara Moto3 2016 Brad Binder, yang pindah ke MotoGP dari KTM. Binder pada awalnya ditempatkan untuk menggantikan Hafizh Syahrin di Red Bull KTM Tech 3. Sebagai gantinya, sesama pembalap Moto2, Iker Lecuona dipromosikan untuk menggantikan Syahrin, yang kembali ke Moto2.[17][27]

Pergantian Tengah Musim
- Stefan Bradl menggantikan Marc Márquez sementara dia menjalani pemulihan dari cedera yang dideritanya pasca kecelakaan di putaran pembukaan.[14]
- Pembalap tes Ducati Michele Pirro menggantikan Francesco Bagnaia di Austria sementara dia pulih dari cedera yang dideritanya dalam kecelakaan selama latihan di putaran Ceko.[10]
- Valentino Rossi dinyatakan positif mengidap virus SARS-CoV-2 pada 15 Oktober, mengharuskan dia untuk karantina minimal 10 hari menurut hukum Italia dan memaksanya untuk melewatkan putaran Aragon dan Teruel. Yamaha mengkonfirmasi bahwa mereka tidak akan menurunkan pengganti Rossi di putaran Teruel pada 25 Oktober.[34][35]
- Pembalap tes Aprilia dan mantan pembalap World Superbike Lorenzo Savadori diumumkan untuk menggantikan Bradley Smith untuk tiga putaran terakhir di Valencia dan Portugal, kemudian menyingkirkan Andrea Iannone pada tahun 2020.[2]
- Pembalap Tech3 Iker Lecuona terpaksa absen dari Grand Prix Eropa di Valencia, karena aturan karantina Andorra untuk kontak dekat dengan individu yang dites positif terkena virus SARS-CoV-2. Saudaranya - dengan siapa ia tinggal di Andorra - dan asisten pribadinya keduanya dinyatakan positif mengidap virus pada 3 November, yang berarti Lecuona dipaksa masuk ke karantina wajib minimal 10 hari. Perubahan Kalender
- Grand Prix Thailand dipindahkan dari putaran ke-15 di 2019 ke putaran ke-2 di 2020.
- Grand Prix Argentina dan Grand Prix Amerika kembali bertukar tempat, dengan Austin menggelar putaran ketiga, sementara Argentina menjadi tuan rumah putaran keempat. Sebelumnya terakhir kali sejak 2015, Austin menggelar putaran kedua, sementara Argentina menjadi tuan rumah putaran ketiga.
