Lexington Avenue Line adalah salah satu jalur divisi IRT di New York City Subway, membentang dari Downtown Brooklyn atau Lower Manhattan ke utara hingga 125th Street di East Harlem.[1] Bentangan di Lower dan Midtown Manhattan adalah bagian dari jalur kereta bawah tanah pertama di New York. Jalur ini dilayani oleh kereta 4 5 6 <6>.
Jalur ini juga dikenal dengan nama IRT East Side Line, karena merupakan satu-satunya jalur di Manhattan yang melayani Upper East Side dan East Midtown secara langsung; jalur empat rel ini adalah jalur angkutan cepat yang paling banyak digunakan di Amerika Serikat. Jumlah penumpang rata-ratanya sebanyak 1,3 juta orang "lebih banyak daripada gabungan jumlah penumpang sistem angkutan cepat San Francisco dan Boston".[2]
Jumlah penumpangnya juga melebihi 798.456 penumpang harian di Washington Metro[3] dan 703.326 penumpang harian di sistem Chicago 'L'. Pembangunan tambahan di IND Second Avenue Line telah dimulai untuk mengurangi kepadatan parah yang diakibatkan jumlah penumpang yang tinggi di Lexington Avenue Line.
Sejumlah stasiun di sepanjang jalur ini telah ditinggalkan. Ketika peron diperpanjang hingga seukuran sepuluh mobil, diusulkan bahwa penutupan stasiun-stasiun tersebut dan pembukaan pintu masuk baru untuk stasiun terdekatnya lebih menguntungkan. Misalnya, 14th Street – Union Square memiliki pintu masuk di 16th Street dan 23rd Street memiliki pintu masuk di 22nd Street, sehingga stasiun 18th Street ditutup karena dekat dengan 14th Street – Union Square dan 23rd Street.
Pada Januari 2010, armada 4 memakai gerbong R142 dan R142A. Armada 5 memakai gerbong R142 dan armada 6 memakai gerbong R142A.
