Tiroid peroksidase (bahasa Inggris: thyroid iodide peroxidase,[1] thyroid peroxidase, iodide peroxidase, thyroperoxidase, TPO, EC 1.11.1.8) adalah protein yang terglikosilasi dan menempel pada membran plasma apisal tirosit,[2] serta merupakan enzim yang berperan dalam sintesis hormon tiroid[3] termasuk hormon T3, dari residu tiroglobulin,[4] di bawah stimulasi TSH.

TPO membentuk homodimer peka tionamida yang merupakan autoantigen universal pada penyakit Graves dan penyakit autoimunitas lain pada kelenjar tiroid[5] dengan memicu autoantibodi IgG dengan daya ikat tinggi.[6] Pada kasus hipertiroidisme, TPO dihambat oleh 6-propyl-2-thiouracil (PTU) atau 1-methyl-2-mercaptoimidazole (MMI).[1]
