The Pentagon, markas Departemen Pertahanan Amerika Serikat

Departemen Pertahanan (Department of Defense, Defense Department, USDOD, DOD, DoD, atau the Pentagon[4]) adalah Departemen Eksekutif Pemerintah Amerika Serikat yang bertugas mengoordinasikan dan mengawasi seluruh lembaga dan fungsi pemerintahan yang berkaitan dengan keamanan nasional dan Angkatan Bersenjata Amerika Serikat. Departemen ini juga merupakan badan dengan karyawan terbanyak di dunia,[5] yaitu lebih dari 2,13 juta tentara, pelaut, marinir, pilot, dan pekerja sipil aktif dan lebih dari 1,1 juta anggota Garda Nasional, Angkatan Udara, Laut, Udara, dan Cadangan Marinir. Jumlah totalnya mencapai 3,2 juta personel, termasuk pekerja sipil.

Departemen yang dipimpin Menteri Pertahanan ini memiliki tiga departemen militer bawahan, Departemen Angkatan Darat A.S., Departemen Angkatan Laut A.S., dan Departemen Angkatan Udara A.S.. Selain itu, ada pula sejumlah Lembaga Pertahanan seperti Defense Advanced Research Projects Agency (DARPA), Defense Intelligence Agency (DIA), Defense Logistics Agency (DLA), Missile Defense Agency, National Geospatial-Intelligence Agency (NGA), National Reconnaissance Office (NRO), National Security Agency (NSA), dan Pentagon Force Protection Agency (PFPA). Semuanya diketuai oleh Menteri Pertahanan. DoD juga mengoperasikan beberapa sekolah dinas gabungan seperti National Defense University (NDU) dan National War College (NWC).

Departemen ini mendapatkan anggaran terbanyak dibandingkan badan federal lainnya. Jumlah anggaran yang diterima mencapai lebih dari satu setengah kali anggaran terikat federal setiap tahunnya.
